目的:探討西南地區(qū)雌激素受體α(estrogen receptor α,ERα)基因多態(tài)性與原性肝癌關(guān)系。方法:選擇西南地區(qū)100名原發(fā)性肝癌患者為實(shí)驗(yàn)組,100名非肝病人群作為對(duì)照組。應(yīng)用分子生物學(xué)的方法分析PvuⅡ,XbaⅠ限制性片段長(zhǎng)度多態(tài)性(restriction fragment length polymorphism,RFLP)。同時(shí)對(duì)人雌激素受體基因上游的短串聯(lián)重復(fù)序列(short tandem repeat,STR)進(jìn)行純化、克隆和序列分析,觀察ERα基因多態(tài)性基因型在實(shí)驗(yàn)組與對(duì)照組中的基因型分布。結(jié)果:PvuⅡ和XbaⅠ限制性片段長(zhǎng)度多態(tài)性在兩組中均呈多態(tài)性分布。病例組TA13等位基因頻率高于對(duì)照組,差異有顯著性,TA15等位基因頻率低于對(duì)照組,差異有顯著性。結(jié)論:ERα基因多態(tài)性與原性肝癌有關(guān),X等位基因可能是其危險(xiǎn)因素,P等位基因可能是其保護(hù)因素,TA13等位基因可能是其危險(xiǎn)因素,TA15等位基因可能是其保護(hù)因素。
