目的:對TGF-β1的G-800A,C-509T,T869C和G915C多態(tài)性位點(diǎn)與IgA腎病及其臨床表現(xiàn)之間的相關(guān)性進(jìn)行了分析。方法:納入119例經(jīng)腎活檢證實(shí)為IgA腎病的患者和116例健康正常成人作為對照組。采用多聚酶鏈?zhǔn)椒磻?yīng)-限制片段長度多態(tài)性分析和多聚酶鏈?zhǔn)椒磻?yīng)-擴(kuò)增受阻突變體系方法測定TGF-β1的G-800A,C-509T,T869C和G915C多態(tài)性位點(diǎn)的基因型,分析其與IgA腎病發(fā)病以及臨床表現(xiàn)的相關(guān)性。結(jié)果:研究對象中G-800A和G+915C這兩個(gè)位點(diǎn)沒有多態(tài)性。IgAN患者C-509T位點(diǎn)的等位基因型分布與正常對照具有顯著性差異(P<0.05),且IgAN患者TT純合子的頻率顯著高于對照組(33% vs 20%,P=0.02)。C869T位點(diǎn)等位基因的分布無顯著性差異。C-509T和C869T位點(diǎn)等位基因分布和患者臨床表現(xiàn)無明顯的相關(guān)性。結(jié)論:TGF-β1基因C-509T位點(diǎn)的T等位基因攜帶可能與IgA腎病的發(fā)病易感性具有相關(guān)性。
