目的:探討Na+,K+-ATP1b1(ATP1b1)對(duì)肝癌細(xì)胞生長(zhǎng)及侵襲能力的影響。方法:將ATP1b1表達(dá)質(zhì)粒ATP1b1-CMV-FLAG轉(zhuǎn)染肝癌細(xì)胞SMMC-7721,以空質(zhì)粒轉(zhuǎn)染作對(duì)照,通過(guò)RT-PCR測(cè)定ATP1b1的mRNA表達(dá),在光鏡下觀察細(xì)胞形態(tài)學(xué)變化,MTT法檢測(cè)細(xì)胞生長(zhǎng)增殖,Transwell細(xì)胞侵襲實(shí)驗(yàn)分析肝癌細(xì)胞的侵襲能力。結(jié)果:ATP1b1-CMV-FLAG轉(zhuǎn)染后,SMMC-7721的ATP1b1 mRNA表達(dá)水平明顯增高(P<0.05),轉(zhuǎn)染ATP1b1-CMV-FLAG組、pFLAG-CMV-1組和脂質(zhì)體組的ATP1b1 mRNA表達(dá)水平分別為1.159、0.182和0.093;轉(zhuǎn)染ATP1b1-CMV-FLAG的SMMC-7721細(xì)胞呈變性改變,其生長(zhǎng)受到抑制,轉(zhuǎn)染相同DNA濃度的ATP1b1-CMV-FLAG組與pFLAG-CMV-1組對(duì)細(xì)胞生長(zhǎng)的抑制效應(yīng)有顯著性差異(P<0.05);Transwell細(xì)胞侵襲實(shí)驗(yàn)顯示轉(zhuǎn)染ATP1b1-CMV-FLAG組的SMMC-7721細(xì)胞的體外侵襲力明顯受抑制(P<0.01)。結(jié)論:實(shí)驗(yàn)結(jié)果說(shuō)明了促進(jìn)ATP1b1表達(dá)能抑制肝癌細(xì)胞的生長(zhǎng)及侵襲行為。
