目的 探討缺氧誘導(dǎo)因子-1α(HIF-1α)與基因系尾型同源盒基因2(CDX2)蛋白在結(jié)直腸腺癌組織中的表達(dá)及其與結(jié)直腸癌發(fā)生、發(fā)展的關(guān)系,并且研究HIF-1α與CDX2表達(dá)之間的相關(guān)性。方法 收集62例結(jié)直腸腺癌及20例正常結(jié)直腸組織標(biāo)本,采用免疫組化SABC法檢測(cè)標(biāo)本中HIF-1α、CDX2蛋白的表達(dá),并采用Spearman等級(jí)相關(guān)分析分析HIF-1α與CDX2表達(dá)之間的相關(guān)性。結(jié)果 在20例正常結(jié)直腸黏膜組織中HIF-1α及CDX2蛋白表達(dá)陽性率分別為5.0%(1/20)及95.0%(19/20),在62例結(jié)直腸腺癌組織中HIF-1α及CDX2蛋白表達(dá)陽性率分別為62.9%(39/62)及69.4%(43/62),不同組織中蛋白表達(dá)的差異均有統(tǒng)計(jì)學(xué)意義(P lt;0.05)。結(jié)直腸腺癌中,HIF-1α、CDX2蛋白的表達(dá)與腫瘤的分化程度、淋巴結(jié)轉(zhuǎn)移及Dukes分期有關(guān)(P lt;0.05); Spearman等級(jí)相關(guān)分析表明,HIF-1α與CDX2的表達(dá)呈負(fù)相關(guān)(r=-0.293 2,P lt;0.05)。結(jié)論 HIF-1α表達(dá)上調(diào)、CDX2表達(dá)下調(diào)可能與結(jié)直腸腺癌的發(fā)生有關(guān),且這兩者間呈負(fù)相關(guān)。HIF-1α可能參與調(diào)控CDX2的表達(dá),進(jìn)而加速了結(jié)直腸癌的惡性進(jìn)展。
