目的 探討腫瘤細(xì)胞裂解物致敏的樹突狀細(xì)胞(DC)作為佐劑對結(jié)腸癌LoVo細(xì)胞系的殺傷作用。
方法 反復(fù)凍融法裂解培養(yǎng)的結(jié)腸癌LoVo細(xì)胞,提取細(xì)胞抗原并致敏DC,然后將后者與自身T淋巴細(xì)胞共同培養(yǎng),觀察其對自身T淋巴細(xì)胞增殖的影響以及活化的T淋巴細(xì)胞對LoVo細(xì)胞的殺傷作用。
結(jié)果 結(jié)腸癌LoVo細(xì)胞裂解物致敏的DC能明顯促進(jìn)T淋巴細(xì)胞的增殖,后者對LoVo細(xì)胞的殺傷作用明顯增強(qiáng)。
結(jié)論 腫瘤細(xì)胞裂解物提取方法簡單,易于臨床實(shí)施,其作為細(xì)胞抗原致敏DC制備的腫瘤疫苗能有效活化并產(chǎn)生腫瘤特異性細(xì)胞毒T淋巴細(xì)胞,將有很好的臨床應(yīng)用前景。
