目的 研究P 物質(zhì)( substance P , SP) 對(duì)體外培養(yǎng)的人低分化胃癌細(xì)胞MKN45 內(nèi)游離鈣濃度的影響。方法 在RPMI 1640 培養(yǎng)液中培養(yǎng)人胃癌細(xì)胞系MKN45 ,應(yīng)用鈣特異性熒光指示劑Furu23/ AM 負(fù)載細(xì)胞,用ASN21377642 (N K21 受體拮抗劑) 、尼卡地平和不同濃度的SP 作用于人胃癌細(xì)胞MKN45 ,采用激光掃描共聚焦顯微鏡觀測胃癌細(xì)胞內(nèi)游離鈣濃度的變化。結(jié)果 在Hanks 液(含鈣離子) 中加入10 、50 及100 nmol/ L SP 可以顯著升高胃癌細(xì)胞內(nèi)鈣離子濃度( P lt; 0. 05) ,并且呈劑量依賴性( P  lt; 0. 05) ; 在無外鈣時(shí),SP 引起細(xì)胞內(nèi)鈣離子濃度升高的幅度低于有外鈣時(shí)( P  lt; 0. 05) ; 在Hanks 液中加入N K21 受體拮抗劑ASN21377642 和鈣通道阻滯劑尼卡地平孵育后,SP 引起胃癌細(xì)胞內(nèi)鈣離子濃度升高的幅度顯著降低,與Hanks 液組比較差異有統(tǒng)計(jì)學(xué)意義( P  lt;0. 05) 。結(jié)論 SP 可以顯著升高胃癌細(xì)胞內(nèi)鈣離子濃度,細(xì)胞內(nèi)鈣離子濃度的升高源于內(nèi)鈣釋放和外鈣內(nèi)流。
