目的 觀察T 淋巴細(xì)胞瘤侵襲轉(zhuǎn)移誘導(dǎo)因子1 ( Tiam 1) 反義寡核苷酸(ASODN) 轉(zhuǎn)染對胃癌細(xì)胞形體重塑的影響。方法 采用層粘連蛋白黏附法,由胃癌MKN245 細(xì)胞株(M0 ) 中篩選獲得高侵襲轉(zhuǎn)移亞株(MH ) 。以脂質(zhì)體介導(dǎo)將Tiam 1 ASODN 轉(zhuǎn)染至MH 細(xì)胞中,并應(yīng)用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT2PCR) 及流式細(xì)胞技術(shù)分別檢測Tiam 1 mRNA 和蛋白的表達(dá)。采用HE 染色、細(xì)胞骨架蛋白染色及掃描電鏡技術(shù)觀察轉(zhuǎn)染與未轉(zhuǎn)染MH 細(xì)胞在形態(tài)學(xué)方面的不同。結(jié)果 與未轉(zhuǎn)染組相比,應(yīng)用0. 43μmol/ L Tiam 1 ASODN 轉(zhuǎn)染可特異性抑制胃癌MH細(xì)胞中Tiam 1 mRNA 和蛋白的表達(dá)( P lt; 0. 01) ; 轉(zhuǎn)染MH 細(xì)胞的膜表面突起及偽足變稀疏或縮短,細(xì)胞骨架結(jié)構(gòu)紊亂程度降低,斑點(diǎn)狀肌動(dòng)蛋白小體減少。結(jié)論 特異性ASODN 轉(zhuǎn)染可有效抑制Tiam 1 在胃癌細(xì)胞中的表達(dá)及胃癌細(xì)胞形體重塑,這可能對胃癌細(xì)胞的侵襲轉(zhuǎn)移產(chǎn)生影響。
