目的 研究金雀異黃素( genistein , Gen) 與52氟尿嘧啶(52fluorouracil ,52FU) 對人胃癌細胞SGC-7901 的抑制作用。方法 采用MTT 法檢測Gen 和5-FU 對SGC-7901 細胞的抑制作用,流式細胞術(shù)檢測細胞周期分布,并在透射電鏡下觀察細胞超微結(jié)構(gòu)改變。結(jié)果 Gen 和5-FU 單用或聯(lián)合應(yīng)用對胃癌SGC27901 細胞的生長均有顯著的增殖抑制作用,且呈劑量和時間依賴性。兩藥聯(lián)合應(yīng)用時,當藥物效應(yīng)在0. 2~0. 8 時,具有協(xié)同或相加作用(CI ≤1) ; 18. 8μmol/ L 的Gen 作用后,62. 97 %的SGC-7901 細胞阻滯在G2 / M 期; 8. 84μmol/ L 的52FU作用后,63. 76 %的SGC-7901 細胞阻滯在S 期; 3. 06 μmol/ L 的Gen 與7. 96 μmol/ L 的5-FU 聯(lián)合應(yīng)用后,67. 46 %的SGC27901 細胞阻滯在G0 / G1 期。Gen 和52FU 均可誘導SGC-7901 細胞凋亡。結(jié)論 Gen 與5-FU 通過阻滯細胞周期和誘導細胞凋亡而抑制胃癌細胞的生長,對胃癌細胞的增殖抑制具有協(xié)同作用。
