目的 探討TLSFJM(JM急性T淋巴細(xì)胞白血病細(xì)胞分泌的抑制因子)作為抑制因子對小腸移植急性排斥反應(yīng)的抑制效應(yīng)及其機(jī)理,并與FK506的抑制效應(yīng)特點(diǎn)和副作用進(jìn)行比較分析.方法 雄性BN和LEW大鼠分別作為供、受體行小腸移植,共分5組: 小腸移植組(SBT組)、大劑量FK506 [0.5 mg/(kg·d)]組、小劑量FK506 [0.25 mg/(kg·d)]組、TLSFJM [10 U/(kg·d)]組和FK506 [0.25 mg/(kg·d)]+TLSFJM [10 U/(kg·d)]組.FK506和TLSFJM分別以肌肉或腹腔注射方式給藥.在不同時(shí)間段分別觀察各組動(dòng)物體重、生存時(shí)間及肝、腎功能,組織病理學(xué)檢查排斥反應(yīng)發(fā)生情況.結(jié)果 TLSFJM應(yīng)用7 d,對移植大鼠肝、腎功能無損害.TLSFJM+小劑量FK506不但避免了大劑量FK506對肝功能的損害,而且顯著延長了受體的生存時(shí)間.但單獨(dú)應(yīng)用TLSFJM仍有一定程度排斥反應(yīng)出現(xiàn),不能明顯延長受體的生存時(shí)間.結(jié)論 TLSFJM作為一種有效的移植免疫抑制因子,聯(lián)合小劑量FK506應(yīng)用能延長宿主和移植腸的生存時(shí)間,延緩排斥反應(yīng)的發(fā)生,減輕排斥反應(yīng)的強(qiáng)度,避免大劑量FK506治療帶來的并發(fā)癥.TLSFJM有望成為一種新型、高效、低毒的免疫抑制劑.
