目的 觀察氯化釓(GdCl3)對(duì)內(nèi)毒素刺激后的小鼠巨噬細(xì)胞來(lái)源的細(xì)胞系RAW264.7細(xì)胞Toll樣受體(TLRs)表達(dá)的影響。方法  RAW264.7細(xì)胞分為空白組、內(nèi)毒素處理組(LPS組)及GdCl3處理組(GdCl3組),采用流式細(xì)胞儀檢測(cè)TLR2/4蛋白表達(dá)情況; 用逆轉(zhuǎn)錄PCR(RT-PCR)法分析細(xì)胞中TLR2/4 mRNA表達(dá)的變化; 用ELISA檢測(cè)細(xì)胞培養(yǎng)上清液中TNF-α的水平。結(jié)果 與LPS組相比,不同濃度的GdCl3作用于RAW264.7細(xì)胞后,其TLR2/4蛋白和基因的表達(dá)以及TNF-α的表達(dá)水平均明顯下降,在觀察濃度范圍內(nèi)以2 000 μmol/L時(shí)最明顯,TLR2/4蛋白: 200 μmol/L時(shí)為(70.2±1.28)%/(66.7±2.59)%,400 μmol/L時(shí)為(64.9±1.43)%/(60.4±1.25)%,2 000 μmol/L時(shí)為(47.4±0.98)%/(32.1±0.74)%,其與 LPS組的(94.4±1.76)%/(95.7±0.87)%比較,P<0.01; TLR2/4 mRNA (A值): 200 μmol/L時(shí)為(76.42±2.76)/(101.72±3.14), 400 μmol/L時(shí)為(75.60±3.76)/(89.65±5.17),2 000 μmol/L時(shí)為(64.22±4.67)/(78.44±4.88),其與 LPS組的(127.64±3.25)/(119.82±5.59)比較,P<0.05, P<0.01; TNF-α: 200 μmol/L時(shí)為 (2 540±77) pg/ml, 400 μmol/L時(shí)為 (2 041±106) pg/ml,2 000 μmol/L時(shí)為(1 020±220) pg/ml ,其與 LPS組的(4 688±127) pg/ml比較,P<0.01。結(jié)論 GdCl3能明顯抑制內(nèi)毒素引起的RAW264.7細(xì)胞Toll樣受體的表達(dá)及相應(yīng)炎癥因子的生成。
