【摘要】目的 初步分析BAG3在胰腺癌抗凋亡及產(chǎn)生耐藥特性中的作用。 方法 通過免疫組化方法檢測術(shù)前化療與未化療患者手術(shù)切除的胰腺癌組織中BAG3的表達(dá)水平; 通過蛋白質(zhì)印跡方法檢測MIACaPa2、 PANC1及SW1990 3株細(xì)胞在化療藥物(5FU 50 μg/ml、 MMC 0.5 μg/ml及EADM 1.5 μg/ml)作用18 h后BAG3蛋白表達(dá)的差異。 結(jié)果 術(shù)前化療患者的胰腺癌組織中BAG3呈中強(qiáng)度陽性表達(dá)者的比率與術(shù)前未化療患者相比有升高趨勢,但差異無統(tǒng)計(jì)學(xué)意義(P gt;0.05)?;熕幬锾幚砗?8 h,3株胰腺癌細(xì)胞中BAG3的表達(dá)水平與未經(jīng)化療藥物處理的對照組相比明顯升高(P<0.05)。 結(jié)論 化療可使胰腺癌細(xì)胞中BAG3的表達(dá)升高,BAG3的上調(diào)可能與胰腺癌對化療產(chǎn)生耐藥相關(guān)。
