【摘要】目的 探討低強度微波(LIM)輻射對血管再狹窄病理過程的影響,以評價LIM輻射防治再狹窄的可能性。 方法 健康雄性新西蘭大白兔48只,隨機分為2組。應(yīng)用Fogarty導(dǎo)管損傷兔髂外動脈(EIA)內(nèi)膜,建立再狹窄動物模型。輻射組采用2 450 MHz微波,功率分別為2、5和10 mW/cm2,局部輻射EIA(1 h/d)。分別于術(shù)后3、7、14及28 d 取材。采用HE和EF染色,觀察EIA形態(tài)學(xué)改變及內(nèi)膜面積、中膜面積及管腔狹窄率; TUNEL標記法檢測細胞凋亡; TEM觀察血管平滑肌細胞(VSMC)表型及超微結(jié)構(gòu)。 結(jié)果 LIM微波輻射后,再狹窄早期炎癥反應(yīng)受到抑制,附壁血栓減少,各時間點VSMC的增殖、遷移被抑制,內(nèi)膜面積和管腔狹窄率顯著低于對照組(P<0.01)。 細胞凋亡率在術(shù)后3 d各組間差異無統(tǒng)計學(xué)意義(P gt;0.05),其余時間點,輻射組顯著高于對照組(P<0.01),其中5 mW/cm2者細胞凋亡率明顯高于其他各組(P<0.01)。TEM觀察到對照組大量合成型VSMC術(shù)后14 d達(93.50+3.45)%,輻射組VSMC以收縮型為主,大量成纖維細胞和VSMC呈凋亡的形態(tài)學(xué)改變。

結(jié)論 LIM輻射能顯著抑制再狹窄病理過程中的早期炎癥反應(yīng)和VSMC的增殖、遷移,促進細胞凋亡,有效抑制再狹窄的發(fā)生、發(fā)展。LIM輻射存在量效關(guān)系和功率窗效應(yīng),功率強度為5 mW/cm2時對再狹窄的效應(yīng)最強。
