目的建立人肝細(xì)胞癌多藥耐藥亞株并對(duì)其主要特征進(jìn)行研究。方法應(yīng)用SMMC7721細(xì)胞株,通過(guò)不斷提高培養(yǎng)液中阿霉素(ADM)的濃度,長(zhǎng)期篩選培養(yǎng),得到了人肝細(xì)胞癌多藥耐藥亞株SMMC7721/ADM后,對(duì)其特征進(jìn)行研究。結(jié)果①SMMC7721/ADM對(duì)ADM的抗藥性提高了33.3倍,對(duì)長(zhǎng)春新堿的抗藥性提高了16.8倍,對(duì)順鉑的抗藥性提高了2.8倍; ②耐藥細(xì)胞與親代細(xì)胞的生長(zhǎng)速度基本相同,倍增時(shí)間分別為32.0 h和30.5 h,呈現(xiàn)相似的生長(zhǎng)曲線; ③利用掃描和透射電鏡發(fā)現(xiàn),耐藥細(xì)胞與親代細(xì)胞形態(tài)上的明顯差別是前者微絨毛變得粗短、稀疏、分布不均; ④耐藥細(xì)胞保持著肝細(xì)胞癌的特征,能在裸小鼠皮下移植再現(xiàn); ⑤SMMC7721/ADM在去藥培養(yǎng)1個(gè)月和2個(gè)月后耐藥指數(shù)下降為28.0%和9.2%,而在ADM(0.04 μg/ml)存在下生長(zhǎng)2個(gè)月后,耐藥倍數(shù)可保持穩(wěn)定(35.4倍)。結(jié)論SMMC7721/ADM細(xì)胞亞株具有穩(wěn)定的耐藥性細(xì)胞株的基本特征。
