目的觀察急性胰腺炎發(fā)生時肝組織中核因子κB(NFκB)對TNFα mRNA表達的調(diào)節(jié)及其在肝損傷中的作用。方法Wistar大鼠72只,隨機均分為急性胰腺炎組(AP組)、急性胰腺炎吡咯基二硫氨基甲酸酯(PDTC)處理組(APP組)以及對照組(SO組)。分別在術(shù)后3 h、6 h、12 h及24 h檢測肝組織NFκB活性、TNFα mRNA的表達以及血漿ALT水平。結(jié)果AP組及APP組的NFκB活性、TNFα mRNA表達及血漿ALT水平分別在術(shù)后3~6 h及3~24 h顯著高于SO組。在應(yīng)用了NFκB抑制劑的APP組,NFκB活性、TNFα mRNA表達以及血漿ALT水平均顯著低于AP組。結(jié)論急性胰腺炎發(fā)生時,肝臟中活化的NFκB促進了TNFα mRNA的表達,并參與了肝損傷的發(fā)生。
