目的探討移植靜脈中細胞凋亡及相關基因表達的動態(tài)變化及意義。方法建立100只Wistar大鼠自體頸靜脈移植于腎下腹主動脈模型。采用原位DNA片斷末端標記(TUNEL)和免疫組織化學法,檢測移植靜脈血管平滑肌細胞(VSMCs)凋亡及相關基因bcl2和bax的表達。結果術后1~8周,移植靜脈凋亡的VSMCs多于對照靜脈(P<0.01); 術后2周為凋亡高峰,其凋亡細胞達(28.5±16.6)%,4~8周逐漸下降至(8.1±2.8)%;與對照靜脈〔(0.5±0.2)%〕相比差異有顯著性意義(P<0.01)。 術后1~2周,bcl2表達增多〔(22.1±5.4)%〕,與對照組〔(4.3±2.1)%〕及4~8周組間比較差異有顯著性意義(P<0.01); 移植后1~6周,bax表達顯著高于對照組〔(5.5±2.3)%〕及術后8周組〔(8.2±2.9)%〕,P<0.01。結論 bcl2與bax基因與移植靜脈VSMCs的凋亡密切相關; VSMCs凋亡是移植靜脈重塑和狹窄的重要因素。
