目的 探討希羅達(Xeloda)對人肝癌組織AFP mRNA表達的影響。
方法 抽提所收集的希羅達治療組及對照組肝癌組織標本總RNA(共41例),采用逆轉錄巢式聚合酶鏈反應(RTnested PCR)技術檢測希羅達治療組及對照組AFP mRNA的表達情況。
結果 以RT-nested PCR擴增的AFP基因片段為101 bp,與原設計一致。在希羅達治療組中,21例肝癌組織標本中有12例AFP mRNA呈陽性表達,陽性率為57.14%; 在對照組中,20例肝癌組織標本中有18例AFP mRNA呈陽性表達,陽性率為90.00%,兩者間差異有顯著性意義(P<0.05)。術后4周,希羅達治療組血清AFP濃度為(23.2±12.8) μg/L,明顯低于對照組的(39.6±24.3) μg/L(P<0.05),而兩者術前血清AFP濃度差異無顯著性意義。
結論 希羅達能有效抑制人肝癌組織AFP mRNA的表達,并降低其表達產(chǎn)物血清AFP濃度。其在治療原發(fā)性肝癌的臨床應用價值值得進一步研究。
