目的 研究碘與自身免疫性甲狀腺炎(EAT)的關系。
方法 用碘和甲狀腺球蛋白(TG)誘導SD大鼠建立甲狀腺炎模型,隨機分為5組,每組10只。正常對照組(NC組); 少量碘組(LI組): 飲500 μg/L碘化鈉水; 大量碘組(HI組): 飲500 mg/L碘化鈉水; TG組: 于實驗第15天大鼠接受皮內多點注射完全福氏佐劑乳化的TG各100 μg作為初次免疫,2周后以不完全福氏佐劑乳化的TG加強免疫,以后每周加強免疫1次至第6周末實驗結束; TG+HI組: TG及HI給藥方法同上。至第6周末實驗結束處死所有動物,用放免法檢測血清TG抗體(TGAb)及甲狀腺過氧化物酶抗體(TPOAb)效價,并于光鏡下觀察甲狀腺組織淋巴細胞浸潤程度。
結果 TG+HI組、HI組及TG組甲狀腺濾泡破壞及淋巴細胞浸潤均較明顯,TG+HI組及HI組與NC組比較,P<0.05,TG組與NC組比較有增高趨勢,但無統(tǒng)計學意義。3組TGAb、TPOAb均明顯高于NC組及LI組(P<0.01; 僅HI組TPOAb P<0.05)。各組TGAb及TPOAb水平與甲狀腺病理分級呈正相關關系(r=0.9,P<0.05)。
結論 高碘可誘發(fā)SD鼠EAT發(fā)生,TG與高碘結合誘導SD大鼠EAT的作用更明顯。
