目的通過檢測(cè)腫瘤壞死因子相關(guān)細(xì)胞凋亡誘導(dǎo)配體受體-4(TRAIL-R4)在胰腺癌組織中的表達(dá),探討胰腺癌細(xì)胞抵抗細(xì)胞凋亡的機(jī)理。方法應(yīng)用mRNA印跡法(Northern blotting)、蛋白質(zhì)印跡法(Western blotting)和免疫組織化學(xué)技術(shù),定性、定位分析TRAIL-R4在正常胰腺組織和胰腺癌組織中的表達(dá)。結(jié)果TRAIL-R4 mRNA和蛋白在正常胰腺組織中呈弱表達(dá)或不表達(dá),而在胰腺癌組織中呈高表達(dá); 免疫組織化學(xué)檢測(cè)顯示,在胰腺癌細(xì)胞中TRAIL-R4蛋白呈強(qiáng)染色。結(jié)論TRAIL-R4表達(dá)水平在正常胰腺組織與胰腺癌組織中存在顯著差異,提示胰腺癌細(xì)胞中可能存在對(duì)TRAIL介導(dǎo)的細(xì)胞凋亡抵抗新機(jī)理。
