的探討一套高效液相色譜電化學(xué)系統(tǒng)檢測(cè)尿8羥基脫氧鳥(niǎo)苷(8OHdG)水平的方法學(xué)。方法采用ESA 5600 Coularray系統(tǒng)檢測(cè)廣西扶綏縣肝癌戶人群21例及南寧市對(duì)照人群63例尿8OHdG水平。結(jié)果8OHdG峰與雜質(zhì)峰分離良好; 8OHdG濃度在0.01~15 ng/ml間呈良好線性; 保留時(shí)間、標(biāo)樣和內(nèi)對(duì)照的重復(fù)性試驗(yàn)的批內(nèi)差異(CV)均小于8%; 標(biāo)樣(F=0.002, P=1.000)和內(nèi)對(duì)照(F=2.965, P=0.082)的重復(fù)性試驗(yàn)的批間差異經(jīng)方差分析均無(wú)顯著性意義。采用上述方法測(cè)得84例總的尿8OHdG水平為(5.26±0.41)ng/mg肌酐,高于同類(lèi)研究的檢測(cè)值〔(4.62±0.091)ng/mg肌酐, n=32〕,但兩地人群間尿8OHdG水平差異無(wú)顯著性意義。結(jié)論這是對(duì)肝癌高發(fā)區(qū)高危人群氧化性DNA損傷的首次報(bào)告。通過(guò)謹(jǐn)慎設(shè)計(jì)試驗(yàn)和優(yōu)化試驗(yàn)參數(shù),該系統(tǒng)對(duì)8OHdG峰的分離優(yōu)于以往報(bào)告,適用于尿8OHdG的高通量定量分析,可應(yīng)用于人群氧化性DNA損傷的研究。
