目的探討重組人生長(zhǎng)激素(rhGH)在體外對(duì)人直腸癌細(xì)胞增殖的影響。方法實(shí)驗(yàn)分為對(duì)照組、rhGH組、奧沙利鉑(LOHP)組和rhGH+LOHP組4組,利用體外細(xì)胞培養(yǎng)、MTT比色技術(shù)及流式細(xì)胞儀等方法,測(cè)定不同濃度的rhGH對(duì)人直腸癌細(xì)胞株HR8348細(xì)胞倍增時(shí)間、細(xì)胞抑制率、細(xì)胞周期、增殖指數(shù)(PI)和DNA抑制率的影響。結(jié)果rhGH在體外不促進(jìn)HR8348細(xì)胞的分裂增殖,rhGH組與對(duì)照組比較,以及rhGH+LOHP組與LOHP組比較,其差異均無(wú)統(tǒng)計(jì)學(xué)意義(P gt;0.05); rhGH+LOHP組與對(duì)照組比較及rhGH+LOHP組與對(duì)應(yīng)的rhGH組配對(duì)比較,細(xì)胞倍增時(shí)間明顯延長(zhǎng),細(xì)胞抑制率增加,阻滯于G0~G1期的細(xì)胞數(shù)增加,S期和G2~M期細(xì)胞明顯減少,PI明顯降低,DNA抑制率顯著升高(P<0.01,S期P<0.05)。結(jié)論rhGH在體外不促進(jìn)直腸癌細(xì)胞的分裂增殖。
