目的用腫瘤壞死因子α(TNFα)基因轉(zhuǎn)導(dǎo)的人肝癌細(xì)胞作為疫苗接種小鼠,研究其對(duì)小鼠肝癌移植瘤的免疫排斥作用。方法取昆明種小鼠40只,分為5組,每組8只。分別采用經(jīng)60Co照射的TNFα基因轉(zhuǎn)導(dǎo)的人肝癌細(xì)胞(BEL7404TNFCo組)、未經(jīng)60Co照射的TNFα基因轉(zhuǎn)導(dǎo)的人肝癌細(xì)胞(BEL7404TNF組)、經(jīng)60Co照射的人肝癌細(xì)胞(BEL7404Co組)以及未經(jīng)60Co照射的人肝癌細(xì)胞(BEL7404組)接種小鼠,生理鹽水對(duì)照組僅注射生理鹽水。然后將小鼠肝癌細(xì)胞株H22移植于上述5組小鼠,觀察肝癌移植瘤生長(zhǎng)情況,并應(yīng)用脫氧核糖核酸末端轉(zhuǎn)移酶介導(dǎo)的缺口末端標(biāo)記(TUNEL)法觀察腫瘤細(xì)胞凋亡指數(shù)。結(jié)果BEL7404TNFCo組小鼠接種經(jīng)60Co照射的TNFα基因轉(zhuǎn)導(dǎo)的人肝癌細(xì)胞疫苗后,肝癌移植瘤成瘤率下降,瘤體生長(zhǎng)受抑制,細(xì)胞凋亡指數(shù)增高,與BEL7404Co組、BEL7404組及生理鹽水對(duì)照組相比,差異有顯著性(P<0.01),但與BEL7404TNF組相比,差異則無統(tǒng)計(jì)學(xué)意義(P gt;0.05)。結(jié)論TNFα基因轉(zhuǎn)導(dǎo)的人肝癌細(xì)胞疫苗對(duì)小鼠肝癌移植瘤有較強(qiáng)的免疫排斥作
用。
