目的探討自主神經(jīng)控制對人肝細胞增殖的影響及其機理,并對增殖相關(guān)受體定位。方法用去甲腎上腺素(NE)及其拮抗劑及激動劑和乙酰膽堿(Ach)及其阻滯劑作用于人傳代正常肝L02細胞及肝癌Bel7402細胞,分別于4 h、24 h、48 h及72 h用改良MTT法檢測上述藥物對細胞增殖的影響; 免疫組織化學(xué)染色法對肝L02細胞的腎上腺素受體(α1BAR及β2AR)和表皮生長因子受體(EGFR)定位。結(jié)果NE對人正常肝細胞及肝癌細胞具有促增殖作用,且隨濃度增加其促增殖作用也增加,4 h的增殖率較其它時間點顯著增高(P<0.05); NE+心得安組的作用與NE組間差異無顯著性意義; 二羥苯基異丙氨基乙醇的作用不明顯; Ach對人正常肝細胞的增殖具有抑制作用,且隨濃度增加其抑制作用也增加,加用阿托品在24 h及48 h與Ach比抑制率顯著降低(P<0.05); 琥珀膽堿組在24 h、48 h及72 h對細胞的增殖也具有抑制作用(P<0.05,P<0.01)。所有人肝L02細胞都呈α1BAR、β2AR及EGFR免疫反應(yīng)陽性,主要定位于胞膜及胞漿。結(jié)論血清存在下,交感神經(jīng)遞質(zhì)NE早期對人正常肝細胞L02及人肝癌細胞Bel7402具有促增殖作用,其作用是通過激動αAR實現(xiàn)的, βAR對人肝細胞增殖無影響。副交感神經(jīng)遞質(zhì)Ach對人正常肝細胞L02的增殖具有抑制作用,其作用是通過激動M及N型膽堿受體實現(xiàn)的。
