目的觀察移植胰腺的腺泡細胞凋亡及其與急性排斥反應(yīng)的關(guān)系。方法選用SD和Wistar大鼠進行全胰十二指腸移植。實驗分為同基因移植組(WistarWistar)和異基因移植組(SDWistar)兩組。于術(shù)后第3 d、5 d和7 d分批處死受體,取移植胰腺標本用HE染色和原位末端標記(TUNEL)技術(shù)檢測移植胰腺切片,進行排斥反應(yīng)的病理學(xué)評分和計數(shù)凋亡指數(shù)(AI)。結(jié)果發(fā)生凋亡的細胞主要是腺泡細胞,同基因移植組胰腺有散在的腺泡細胞凋亡,AI在術(shù)后無明顯變化。異基因移植組胰腺腺泡細胞凋亡在術(shù)后第3 d、5 d和7 d逐漸升高,AI與急性排斥反應(yīng)的病理學(xué)評分成正相關(guān)。結(jié)論細胞凋亡與移植胰腺急性排斥反應(yīng)的嚴重程度顯著相關(guān),凋亡指數(shù)可作為判斷移植物損傷程度的指標,對急性排斥反應(yīng)的診斷有一定的參考價值。
