目的 探討槲皮素抑制乳腺癌發(fā)生及增殖的作用。方法 建立二甲基苯丙蒽(DMBA)誘導(dǎo)的乳腺癌動(dòng)物模型。79只雌性SD大鼠隨機(jī)分為A組(DMBA)、B組〔DMBA+三苯氧胺(TAM)〕、C組(DMBA+槲皮素)及D組(空白對(duì)照)。持續(xù)喂養(yǎng)28周,經(jīng)光鏡、電鏡觀察,抗PCNA及Hras免疫組化分析。結(jié)果 ①A組大鼠乳腺腫瘤發(fā)生率為76.2%,明顯高于B組(40.9%)、C組(45.5%)及D組(0%),P<0.05; B組與C組相比差異無(wú)顯著性意義(P>0.05)。②A組大鼠乳腺癌平均直徑2.37cm,明顯大于B組(1.82cm)及C組(1.71cm),P<0.05; B組和C組相比差異無(wú)顯著性意義(P>0.05)。③PCNA免疫組化染色顯示: A組與B組和C組相比差異有顯著性意義(P<0.05),B組和C組相比差異無(wú)顯著性意義(P>0.05)。④Hras免疫組化染色顯示: A組與B組和C組比較差異有顯著性意義(P<0.05),B組和C組相比差異無(wú)顯著性意義(P>0.05)。結(jié)論 槲皮素有降低DMBA誘導(dǎo)的SD大鼠乳腺癌發(fā)生率及抑制腫瘤生長(zhǎng)的作用,其機(jī)理可能與抑制ras基因活性、阻抑細(xì)胞增殖有關(guān)。
