目的  探討鈣離子對肝臟的損傷作用。方法  應(yīng)用含膠原酶保存液灌注大鼠肝臟制成不同細胞成活率的肝細胞懸液,低溫保存肝臟細胞。應(yīng)用Fura-2/AM標記測細胞內(nèi)鈣。 結(jié)果  ①低溫保存2小時肝細胞: 實驗1組(細胞成活率為5%)肝細胞內(nèi)鈣值為(1055.0±30.79) nmol/L; 實驗2組(細胞成活率為10%)肝細胞內(nèi)鈣值為(853.0±20.42) nmol/L; 實驗3組(細胞成活率為30%)肝細胞內(nèi)鈣值為(616.0±13.20) nmol/L; 實驗4組(細胞成活率為50%)肝細胞內(nèi)鈣值為(562.0±26.06) nmol/L; 實驗5組(細胞成活率為70%~80%)肝細胞內(nèi)鈣值為(318.0±13.01) nmol/L; 實驗6組(細胞成活率為90%)肝細胞內(nèi)鈣值為(114.6±6.11) nmol/L。②低溫保存24小時肝細胞: 實驗A組(細胞成活率為10%)肝細胞內(nèi)鈣值為(1704.0±70.11) nmol/L; 實驗B組(細胞成活率為50%)肝細胞內(nèi)鈣值為(1125.0±23.22) nmol/L。隨著細胞成活率的降低,細胞內(nèi)鈣值逐漸升高; 在同樣細胞成活率情況下,隨著保存時間延長(2小時與24小時)細胞內(nèi)鈣值增高1倍左右。結(jié)論  在低溫保存過程中,鈣超載是造成肝臟細胞缺血再灌注損傷的一個主要因素。
