為觀察灌注壓力對肝移植物活性的影響,實驗選擇SD大鼠,經門靜脈在不同壓力下灌注4℃的Collin’s液以保存肝臟,然后應用高效液相色譜儀檢測各組的高能磷酸化合物,制備出肝細胞懸液后采用陽離子測定儀測定肝細胞內游離鈣濃度,并同時觀察了肝臟的超微結構變化及光鏡病理改變。結果發(fā)現(xiàn):隨灌注壓力的升高,肝臟的能量代謝、肝細胞內游離鈣濃度以及超微結構均有明顯改變。本實驗結果提示:不適當?shù)墓嘧毫蓪е赂闻K移植物的能量代謝的變化及肝細胞的鈣超載,從而影響肝移植物的活力; 為了取得高質量的供肝,應采用低溫低壓灌注。
