為研究肝細(xì)胞低密度脂蛋白受體對(duì)膽汁中膽汁酸的影響及其在膽囊結(jié)石過(guò)程中可能的機(jī)理,采用高膽固醇膳食誘發(fā)兔膽囊結(jié)石模型,對(duì)進(jìn)食后1、2、3、4周組及對(duì)照組動(dòng)物,采用雙波長(zhǎng)薄層掃描法測(cè)定膽汁中膽汁酸,125I-LDL放免標(biāo)記法測(cè)定肝細(xì)胞低密度脂蛋白受體(LDLr)活性。結(jié)果: 高膽固醇膳食后2、3、4周組分別有4/10,6/10,7/10出現(xiàn)膽囊結(jié)石; 膽汁中甘氨膽酸明顯降低(P<0.05);125I-LDL與肝細(xì)胞低密度脂蛋白受體(LDLr)的最大結(jié)合力(Bmax)逐漸下降(P<0.05),解離常數(shù)(Kd)值逐漸升高(P<0.05)。上述結(jié)果提示: 肝細(xì)胞低密度脂蛋白受體活性下降,可能導(dǎo)致膽汁中甘氨膽酸減少,致成石性膽汁形成。
