用裸小鼠胃癌細(xì)胞移植瘤作為體內(nèi)誘導(dǎo)分化研究模型,采用移植瘤生長(zhǎng)狀態(tài)、移植瘤組織學(xué)、超微結(jié)構(gòu)、細(xì)胞周期動(dòng)力學(xué)變化及移植瘤p53、p21蛋白表達(dá)作為指標(biāo),研究了不同劑量的全反式維甲酸(ATRA)對(duì)裸小鼠胃癌細(xì)胞移植瘤的抗增殖及誘導(dǎo)分化作用。結(jié)果發(fā)現(xiàn):在ATRA 300~1 000μg/只·d的劑量下,對(duì)移植瘤具有明顯生長(zhǎng)抑制作用,并對(duì)移植瘤細(xì)胞有較強(qiáng)的誘導(dǎo)分化作用;同時(shí)可抑制移植瘤癌細(xì)胞p53、p21的表達(dá)。本實(shí)驗(yàn)結(jié)果提示:ATRA在體內(nèi)對(duì)人胃癌細(xì)胞有較強(qiáng)的抗增殖和誘導(dǎo)分化作用,這種抗癌作用可能與ATRA抑制癌細(xì)胞p53及p21表達(dá),促進(jìn)癌細(xì)胞凋亡有密切關(guān)系。
