對(duì)25例大腸癌根治術(shù)患者的新鮮標(biāo)本進(jìn)行細(xì)胞分離和原代培養(yǎng)，采用噻唑氮藍(lán)(MTT)比色分析法和氚-胸腺嘧啶核苷(3H-TdR)摻入法，在體外觀察5肽胃泌素(PG)對(duì)原代培養(yǎng)大腸癌活細(xì)胞數(shù)(吸光度，Α值)和DNA合成(脈沖數(shù)，CPM值)的影響。結(jié)果：PG組高、中及低分化腺癌活細(xì)胞數(shù)和DNA合成均明顯高于對(duì)照組(Ｐ＜0.01，Ｐ＜0.05)。出現(xiàn)明顯促細(xì)胞生長所需PG濃度高分化腺癌組為0.3906μg/ml，而中、低分化腺癌組均為6.2500μg/ml。本研究結(jié)果提示：胃泌素可促進(jìn)人大腸癌細(xì)胞的增殖，尤以高分化腺癌對(duì)胃泌素更敏感。這為大腸癌患者特別是高分化腺癌應(yīng)用胃泌素受體拮抗劑進(jìn)行內(nèi)分泌治療提供了實(shí)驗(yàn)依據(jù)。
