目的  建立大鼠銅綠假單胞菌(PA)慢性肺部感染模型。方法  健康Sprague-Dawley大鼠分為銅綠假單胞菌感染組與對(duì)照組。將銅綠假單胞菌包被的硅膠管(3Fr,直徑1.0 mm,長(zhǎng)3.0 mm)置入感染組大鼠的一側(cè)主支氣管處;對(duì)照組置入生理鹽水浸泡的無(wú)菌硅膠管。接種后28 d處死大鼠,分離肺組織,進(jìn)行病理學(xué)檢查和肺組織勻漿菌落計(jì)數(shù)。結(jié)果  硅膠管置入后28 d,感染組肺組織培養(yǎng)出綠色的野生型銅綠假單胞菌,勻漿菌落計(jì)數(shù)超過(guò)103cfu/g;大體病理表現(xiàn)為纖維增生、肉芽腫形成,鏡下表現(xiàn)為淋巴細(xì)胞浸潤(rùn)為主炎癥反應(yīng);對(duì)照組大鼠肺組織未培養(yǎng)出銅綠假單胞菌,病理上未見(jiàn)明顯改變。結(jié)論  銅綠假單胞菌包被的硅膠管置入大鼠一側(cè)主支氣管,可成功建立慢性肺部感染模型。該方法造價(jià)低廉、可操作性較強(qiáng),易于推廣,為PA肺部感染藥物治療提供了依據(jù)。
