目的  通過建立大鼠肺癌模型,研究山葵提取液對肺癌早期分子標(biāo)志物hnRNP A2/B1的調(diào)節(jié)作用,進(jìn)一步闡明山葵抗肺癌作用的分子機制。方法  將36只Wistar大鼠隨機分為肺癌模型組(對照組)和山葵治療組(實驗組)。按1 mL碘油內(nèi)含3-甲基膽蒽(MCA)50 mg配制致癌質(zhì)碘油。每只大鼠均用MCA碘油溶液0.1 mL在左肺葉支氣管內(nèi)灌注誘癌。實驗組于誘癌前1周開始胃飼山葵提取液至動物被處死,對照組于同一時間胃飼等量生理鹽水。造模后第30、60及90 d分批處死大鼠,每次6只。免疫組織化學(xué)法和RT-PCR法檢測肺癌組織hnRNP A2/B1的蛋白和基因表達(dá)。結(jié)果  山葵提取液能降低肺癌組織hnRNP A2/B1的蛋白表達(dá),在第30,60及90 d的抑制率分別為51.0%,51.0%和45.1%。山葵提取液同時降低了hnRNP A2/B1 mRNA的表達(dá),在第60和90 d的抑制率分別為79.5%和58.0%。結(jié)論  山葵提取液能降低肺癌早期分子標(biāo)志物hnRNP A2/B1的表達(dá),可能是其抗肺癌的分子機制之一。
