目的  探討水通道蛋白1(AQP-1)在炎癥性胸腔積液大鼠胸膜上的表達(dá)及其與胸水形成的關(guān)系。方法  56只健康Wistar大鼠隨機(jī)分為對(duì)照組(8只)和胸膜炎組(48只)。采用角叉菜膠胸腔內(nèi)注射復(fù)制炎癥性胸腔積液大鼠模型。胸膜炎組根據(jù)注入角叉菜膠后處理時(shí)間再分為6,12,24,36,48和72 h組,每組8只。測(cè)定各組大鼠胸腔積液量,對(duì)胸腔積液進(jìn)行細(xì)胞計(jì)數(shù)和分類(lèi),采用HE染色觀察臟、壁層胸膜的病理變化,采用免疫組化染色檢測(cè)AQP-1在大鼠胸膜上的分布,分別采用RT-PCR和Western blot方法檢測(cè)臟、壁層胸膜AQP-1的mRNA和蛋白表達(dá)。結(jié)果  胸膜炎各時(shí)相組胸水量分別為2.10±0.22、4.10±0.15、4.40±0.36、3.20±0.27、2.60±0.18和0.12±0.02 mL。AQP-1主要表達(dá)在臟、壁層胸膜的間皮細(xì)胞和毛細(xì)血管內(nèi)皮細(xì)胞。胸膜炎各時(shí)相組臟、壁層胸膜AQP-1表達(dá)均增強(qiáng)。胸膜炎組AQP-1 mRNA在壁層胸膜的表達(dá)于6 h開(kāi)始升高,24 h達(dá)高峰;在臟層胸膜的表達(dá)于12 h開(kāi)始升高,24 h達(dá)高峰。AQP-1蛋白表達(dá)的變化趨勢(shì)與mRNA表達(dá)一致。12 h和24 h組壁層胸膜AQP-1表達(dá)與胸腔積液量正相關(guān)(r=0.857,r=0.846,P均 lt;0.01);24 h臟層胸膜AQP-1表達(dá)與胸腔積液量正相關(guān)(r=0.725,P lt;0.05)。結(jié)論  胸膜炎胸腔積液時(shí)胸膜AQP-1表達(dá)增強(qiáng),AQP-1可能參與炎癥性胸腔積液的轉(zhuǎn)運(yùn)過(guò)程。
