目的  觀察低氧習(xí)服對模擬高原肺水腫大鼠肺組織結(jié)構(gòu)及低氧誘導(dǎo)因子1α(HIF-1α)表達的影響。方法  56只Wistar大鼠隨機分為7組:常氧對照組(N組)、急性低氧對照組(H0組)、急性低氧組(H1組)、3 000 m低氧習(xí)服組(C3.0組)、3 000 m低氧習(xí)服+急性低氧組(C3.1組)、5 000 m低氧習(xí)服組(C5.0組)、5 000 m低氧習(xí)服+急性低氧組(C5.1組),每組8只。習(xí)服完成后,將大鼠置于模擬海拔6 000 m急性低氧24 h。采用Western blot和RT-PCR方法檢測動物習(xí)服完成后肺組織HIF-1α蛋白和mRNA的表達情況,光鏡和電鏡下觀察大鼠急性低氧后肺結(jié)構(gòu)變化。結(jié)果  H1組肺組織顯微和超微結(jié)構(gòu)出現(xiàn)明顯間質(zhì)性肺水腫,低氧習(xí)服后間質(zhì)性肺水腫明顯減輕。Western blot方法未檢測到大鼠肺組織表達HIF-1α,但HIF-1α mRNA在N組有一定量表達,C3.0組和C5.0組HIF-1α? mRNA表達均明顯高于N組(P均 lt;0.01)。結(jié)論  低氧習(xí)服可以減輕低氧大鼠肺間質(zhì)水腫程度,提高肺組織HIF-1α mRNA表達。HIF-1可能介導(dǎo)參與了大鼠肺低氧習(xí)服過程。
