目的 探討半夏提取物( EP) 對脂多糖誘導(dǎo)的大鼠氣道黏液高分泌模型的干預(yù)作用。方法 30 只Wistar 大鼠隨機(jī)分為5 組( n= 6) , 分別為空白組、模型組、低劑量EP 組、中劑量EP 組及高劑量EP 組。模型組和EP 組經(jīng)氣道注入脂多糖( LPS) 復(fù)制大鼠氣道黏液高分泌模型。低、中、高劑量EP 組連續(xù)4 d 分別給予管飼EP 10、30 及60 g/kg; 采用免疫組化法檢測肺內(nèi)氣道的黏蛋白5AC ( MUC5AC) 蛋白表達(dá), RT-PCR 法檢測肺組織MUC5AC mRNA、水通道蛋白5( AQP-5) mRNA; ELISA 法檢測BALF 中的TNF-α濃度。結(jié)果 大鼠氣道上皮MUC5AC 蛋白和肺組織MUC5AC mRNA 的表達(dá)在模型組中均明顯高于空白組( P lt; 0. 05) ; 在低劑量及中劑量EP 組較模型組稍降低( P gt;0. 05) , 仍顯著高于空白組( P lt; 0. 05) ; 在高劑量EP 組顯著低于模型組( P lt;0. 05) 。大鼠AQP-5 mRNA 表達(dá)在模型組顯著低于空白組( P lt;0. 05) ; 在低劑量及中劑量EP 組較模型組稍升高( P gt; 0. 05) , 仍較空白組顯著降低( P lt;0. 05) ; 在高劑量EP 組顯著高于模型組( P lt; 0. 01) 。BALF 中TNF-α濃度與肺組織MUC5AC mRNA 的表達(dá)呈正相關(guān)( r = 0. 948, P lt;0. 05) , 肺組織AQP-5 mRNA 與MUC5AC mRNA 及 TNF-α的表達(dá)呈負(fù)相關(guān)( r = - 0. 955, P lt;0. 05; r = - 0. 909, P lt;0. 05) 。結(jié)論 高劑量的EP 能明顯抑制大鼠氣道上黏液高分泌狀態(tài)。
