目的  通過建立煙曲霉菌絲感染人臍靜脈內(nèi)皮細(xì)胞( HUVEC) 的體外模型, 觀察煙曲霉菌絲感染后HUVEC 表達(dá)血管假性血友病因子( vWF) 的變化, 以及用不同干預(yù)因素處理對(duì)HUVEC表達(dá)vWF 的影響, 以探討侵襲性曲霉病血管侵襲的機(jī)制。方法  實(shí)驗(yàn)分六組, 即空白對(duì)照組、TNF-α組、煙曲霉菌絲組、細(xì)胞松弛素D組、N-鈣黏蛋白抗體組和地塞米松組, 分別于2、6、12 及18 h 提取細(xì)胞培養(yǎng)上清液, 采用ELISA 法檢測(cè)vWF濃度。結(jié)果  與2 h 比較, 空白對(duì)照組和TNF-α組的vWF表達(dá)量在18 h 增加( P  lt;0. 05) , 其余各組vWF表達(dá)量在6、12 及18 h 均增加( P  lt;0. 05) ; 各實(shí)驗(yàn)組與空白對(duì)照組比較, 除N-鈣黏蛋白抗體組HUVEC 的vWF表達(dá)量在2 h無明顯變化外, 其余各時(shí)間點(diǎn)vWF表達(dá)量均升高( P  lt;0. 05) 。TNF-α組HUVEC 的vWF 表達(dá)量在2 h 高于煙曲霉菌絲組, 而在18 h 低于煙曲霉菌絲組。N-鈣黏蛋白抗體組HUVEC 的vWF 表達(dá)量在2 h 和6 h 分別低于煙曲霉菌絲組相應(yīng)時(shí)間點(diǎn)。細(xì)胞松弛素D 組和地塞米松組vWF 在各時(shí)間點(diǎn)與煙曲霉菌絲組的差異均無統(tǒng)計(jì)學(xué)意義。結(jié)論  煙曲霉菌絲感染后, HUVEC 過度表達(dá)vWF, N-鈣黏蛋白單克隆抗體可減少vWF 的表達(dá), 而細(xì)胞松弛素D 和地塞米松對(duì)vWF的表達(dá)無明顯影響。
