目的:探討雌激素饑餓對(duì)腫瘤壞死因子相關(guān)凋亡誘導(dǎo)配體(TRAIL)誘導(dǎo)乳腺癌細(xì)胞MCF-7凋亡的影響及作用機(jī)制?!糎TH〗方法〖HTSS〗:MCF-7細(xì)胞培養(yǎng)貼壁之后,用雌激素饑餓處理后加入TRAIL,用熒光染料Hoechst染色法檢測(cè)細(xì)胞的凋亡,用細(xì)胞計(jì)數(shù)法檢測(cè)細(xì)胞的存活。在雌激素饑餓處理MCF-7細(xì)胞后,收集對(duì)照和饑餓組蛋白用Western blot法檢測(cè)相關(guān)的蛋白表達(dá)?!糎TH〗結(jié)果〖HTSS〗:單獨(dú)使用雌激素饑餓處理或者單獨(dú)使用TRAIL處理乳腺癌細(xì)胞MCF-7都能誘導(dǎo)細(xì)胞凋亡,但是它們誘導(dǎo)凋亡的活性較小,兩種方法聯(lián)合使用可以極大地增加誘導(dǎo)細(xì)胞凋亡的活性(P lt;0.001)。雌激素饑餓處理后的乳腺癌細(xì)胞,死亡受體5(DR5)表達(dá)上調(diào)?!糎TH〗結(jié)論〖HTSS〗:乳腺癌細(xì)胞MCF-7對(duì)TRAIL敏感度不高,雌激素饑餓可以增加TRAIL誘導(dǎo)乳腺癌細(xì)胞凋亡的活性,DR5與雌激素饑餓誘導(dǎo)TRAIL活性增加相關(guān)。
