目的:探討人凋亡相關(guān)新基因PNAS-4(hPNAS-4)基因通過(guò)脂質(zhì)體轉(zhuǎn)染至Lewis肺癌LL2細(xì)胞后對(duì)放射治療的增敏作用?!糎TH〗方法〖HTSS〗:用脂質(zhì)體介導(dǎo)的轉(zhuǎn)染技術(shù),將hPNAS-4基因?qū)隠ewis肺癌LL2細(xì)胞中,Western blot鑒定其過(guò)表達(dá)后,觀察X射線照射對(duì)其集落形成的影響;流式細(xì)胞儀檢測(cè)hPNAS-4基因或/和放療(0,1,2,4,6 Gy)對(duì)LL2細(xì)胞生長(zhǎng)抑制及凋亡的影響?!糎TH〗結(jié)果〖HTSS〗:通過(guò)Western blot證實(shí)了hPNAS-4基因在LL2細(xì)胞中的過(guò)表達(dá)。Lip-hPNAS-4加放射治療處理組細(xì)胞的生存能力明顯降低,腫瘤細(xì)胞的克隆形成明顯減少,流式細(xì)胞術(shù)檢測(cè)體外培養(yǎng)的腫瘤細(xì)胞凋亡明顯增加?!糎TH〗結(jié)論〖HTSS〗:hPNAS-4基因聯(lián)合放射治療能產(chǎn)生協(xié)同抗腫瘤效應(yīng)。
