目的探討預輸注5-FU處理過的同種異基因淋巴細胞對大鼠移植肝的影響。方法實驗分4組,均行Wistar大鼠至SD大鼠的肝移植。 對照組: 僅行肝移植; 單純淋巴細胞組: 受體于術前第7天及第4天各輸注Wistar大鼠淋巴細胞(5×10 6個/ml)懸液1 ml; 低濃度5-FU預處理組: 受體于術前第7天及第4天各輸注經(jīng)7.5 μg 5-FU處理過的Wistar大鼠淋巴細胞(5×10 6個/ml)懸液1 ml; 高濃度5-FU預處理組: 受體于術前第7天及第4天各輸注經(jīng)15 μg 5-FU處理過的Wistar大鼠淋巴細胞(5×10 6個/ml)懸液1 ml。于術后第7天處死SD大鼠,行移植肝臟Fas-L及CD8免疫組化檢測。結(jié)果肝小葉及匯管區(qū)中,低濃度5-FU預處理組Fas-L表達陽性細胞的IOD均高于對照組、單純淋巴細胞組和高濃度5-FU預處理組(P lt;0.05); 單純淋巴細胞組與高濃度5-FU預處理組比較差異無統(tǒng)計學意義(P gt;0.05)。肝小葉中, CD8+ T淋巴細胞的IOD在單純淋巴細胞組、低濃度5-FU預處理組和高濃度5-FU預處理組的差異無統(tǒng)計學意義(P gt;0.05); 在匯管區(qū),低濃度5-FU預處理組的CD8+ T淋巴細胞IOD低于單純淋巴細胞組、高濃度5-FU預處理組和對照組(P lt;0.05)。結(jié)論預輸注經(jīng)低濃度5-FU處理的同種異基因淋巴細胞可以較好地誘導大鼠肝移植免疫耐受,其可能的機理是通過誘導移植物中Fas-L表達的上調(diào),從而獲得免疫耐受。
