目的  建立人中分化結(jié)腸腺癌細(xì)胞裸鼠皮下移植瘤模型,以探討碘-125(125I)粒子治療中分化結(jié)腸腺癌的有效性。
方法  將人中分化結(jié)腸腺癌細(xì)胞(SW480)種植到雄性BALB/c-nu/nu裸小鼠皮下,建立動物模型,用隨機(jī)抽簽法隨機(jī)將荷瘤裸小鼠分成對照組和實(shí)驗(yàn)組(每組24只),對照組植入1顆空白粒子,實(shí)驗(yàn)組植入1顆劑量為1.48×107 Bq的125I粒子,觀察粒子植入后瘤體生長情況,分別于植入后第7、14、21及28天處死實(shí)驗(yàn)組與對照組荷瘤裸小鼠,每個時相6只裸小鼠。剝?nèi)×鲶w,分別用于免疫組化SP法測定增殖細(xì)胞核抗原(PCNA)的表達(dá),原位末端標(biāo)記(TUNEL)法檢測腫瘤細(xì)胞凋亡情況。
結(jié)果  隨著放射時間的延長,從第10天起實(shí)驗(yàn)組腫瘤體積明顯小于對照組(P lt;0.05),從第14天開始PCNA標(biāo)記指數(shù)表達(dá)顯著低于對照組(P lt;0.05),從第21天開始凋亡指數(shù)顯著高于對照組(P lt;0.05)。
結(jié)論   125I粒子通過持續(xù)低劑量輻射后可以下調(diào)PCNA的表達(dá),誘導(dǎo)人中分化結(jié)腸腺癌細(xì)胞凋亡增加,是其抑制人中分化結(jié)腸腺癌增殖可能的生物學(xué)機(jī)理之一。
