目的 探討抗腫瘤相關(guān)糖蛋白-72(TAG-72)單鏈抗體的原核表達(dá)及與4種肝癌細(xì)胞系和肝癌組織的特異性親和力。
方法 將抗TAG-72單鏈抗體的cDNA片段插入噬菌粒載體pCANTAB5E, 獲得噬菌粒載體TAG-72-scFv-pCANTAB5E。將后者轉(zhuǎn)化入E.coli HB2151, 經(jīng)β, D異丙基硫代半乳糖苷(IPTG)誘導(dǎo)表達(dá)獲得抗TAG-72單鏈抗體蛋白。采用細(xì)胞培養(yǎng)及免疫細(xì)胞/組織化學(xué)方法, 檢測4種肝癌細(xì)胞系及石蠟包埋的肝癌組織中TAG-72抗原的表達(dá)。
結(jié)果 SDS-PAGE及Western blot法證實, 抗TAG-72單鏈抗體蛋白分子得以正確表達(dá)。抗TAG-72單鏈抗體可與肝癌細(xì)胞系SMMC7721、HepG2和HHCC結(jié)合, 表明這3種細(xì)胞表達(dá)了特異性腫瘤抗原; 抗TAG-72單鏈抗體不能結(jié)合BEL7402細(xì)胞。40例肝癌組織中TAG-72抗原表達(dá)陽性率Ⅰ期、Ⅱ~Ⅲ期分別為23.08%(3/13)和62.96%(17/27),在正常肝組織標(biāo)本中無TAG-72抗原表達(dá),TAG-72抗原在正常肝組織及肝癌組織中表達(dá)的差異有統(tǒng)計學(xué)意義(P lt;0.05)。
結(jié)論 TAG-72抗原在肝癌細(xì)胞或肝癌組織中有特異性表達(dá)，有可能成為判斷肝癌的標(biāo)志物。
