目的  建立生長激素促分泌素受體(ghrelin receptor,GHS-R)基因敲除小鼠胚胎干細(xì)胞(ES細(xì)胞)雜合子模型,為研究GHS-R基因的功能奠定基礎(chǔ)。
方法  用TK-neo置換原X-pPNT載體的PGK-neo構(gòu)建目標(biāo)載體。以小鼠基因組DNA為模板, 用PCR方法擴(kuò)增2條同源臂,將其按照一定方向裝入含有TK-neo的X-pPNT載體,并測序鑒定。載體線性化及純化后電穿孔轉(zhuǎn)染小鼠ES細(xì)胞,用G418和更昔洛韋(Gancyclovir)對電穿孔轉(zhuǎn)染后的ES細(xì)胞進(jìn)行正、負(fù)篩選培養(yǎng),得到雙藥抗性ES細(xì)胞,克隆后抽提基因組DNA,分別用PCR方法鑒定2條同源臂,并測序確定成功同源重組的ES細(xì)胞克隆。
結(jié)果  改建X-pPNT載體成功,PCR獲得2條同源臂片段,測序正確,并按一定方向裝入打靶載體,ES細(xì)胞轉(zhuǎn)染后經(jīng)雙藥篩選得到328個(gè)陽性ES細(xì)胞克隆,PCR及測序鑒定證實(shí)3個(gè)克隆發(fā)生同源重組。
結(jié)論  本研究成功獲得了GHS-R(-/+)雜合子小鼠ES細(xì)胞克隆,為進(jìn)一步通過顯微注射及雜交育種獲得GHS-R基因敲除小鼠打下了基礎(chǔ)。
