目的了解上皮型鈣黏附分子(E-cadherin)下調(diào)對腫瘤細胞浸潤能力的影響。
方法選用人胰腺癌細胞株JHP-1,經(jīng)E-cadherin反義寡核苷酸(ASODN)作用,應用免疫細胞化學染色、Western blot法檢測細胞E-cadherin蛋白表達情況,細胞浸潤MTT比色法檢測該細胞對基底膜的浸潤能力。
結(jié)果經(jīng)E-cadherin ASODN作用后, JHP-1細胞膜E-cadherin表達減弱, E-cadherin蛋白含量與對照組相比明顯降低; JHP-1細胞對基底膜浸潤能力明顯高于對照組(P<0.001)。
結(jié)論E-cadherin與腫瘤細胞的浸潤能力有關(guān)。
