目的觀察內(nèi)源性光動(dòng)力療法(photodynamic therapy, PDT)對(duì)人結(jié)腸癌SW480細(xì)胞環(huán)磷酸腺苷(cAMP)和環(huán)磷酸鳥苷(cGMP)的影響。
方法將SW480細(xì)胞分為4組: 對(duì)照組、 光照組、 δ氨基酮戊酸 (ALA)組、 內(nèi)源性PDT(ALAPDT)組,用放射免疫法檢測(cè)光照后30、60、90及120 min各組細(xì)胞的cAMP和cGMP的濃度。
結(jié)果cAMP濃度在監(jiān)測(cè)時(shí)間內(nèi),ALAPDT組在30 min時(shí)最高,明顯高于其余各組相應(yīng)時(shí)相及同一組內(nèi)其余時(shí)相(P均<0.001); 在60、90及120 min時(shí)與其它各組相應(yīng)時(shí)相比較,差異均無統(tǒng)計(jì)學(xué)意義; 其余各組在監(jiān)測(cè)時(shí)間范圍內(nèi),組間及組內(nèi)不同時(shí)相間差異均無統(tǒng)計(jì)學(xué)意義。cGMP濃度在各組細(xì)胞的組間及同組內(nèi)不同時(shí)相間差異均無統(tǒng)計(jì)學(xué)意義。
結(jié)論內(nèi)源性PDT在殺傷腫瘤細(xì)胞的同時(shí),SW480細(xì)胞也能通過細(xì)胞內(nèi)cAMP濃度的瞬時(shí)升高產(chǎn)生保護(hù)作用,抑制細(xì)胞凋亡。
