【摘要】目的探討細菌性腹膜炎致多器官功能不全綜合征(MODS)大鼠結腸運動的變化和細胞因子白介素6(IL6)、腫瘤壞死因子α(TNFα)及誘生性一氧化氮合成酶(iNOS)對結腸運動的影響。
方法將Wistar大鼠隨機分成正常對照組和MODS組; 分別觀測2組大鼠排便的糞點數(shù)、結腸肌條的振幅、結腸平滑肌細胞長度以及血清一氧化氮(NO)的變化; 利用免疫組化和逆轉錄聚合酶鏈反應(RTPCR)方法檢測結腸平滑肌IL6、TNFα和iNOS表達的變化。
結果MODS組排便的糞點數(shù)和肌條的振幅均明顯低于正常對照組(P<0.05); IL6、TNFα和iNOS蛋白表達在正常對照組為陰性表達,在MODS組為陽性表達; 在正常對照組未檢出IL6 mRNA、TNFα mRNA和iNOS mRNA的表達,而在MODS組,IL6 mRNA、TNFα mRNA和iNOS mRNA均呈陽性表達; MODS組平滑肌細胞的長度和血清NO水平均較正常對照組明顯增高(P<0.01)。
結論MODS后結腸運動功能障礙與iNOS、IL6和TNFα密切相關。
