目的研究survivin反義寡核苷酸(ASODN)對膽囊癌細(xì)胞凋亡的誘導(dǎo)作用。
方法采用脂質(zhì)體介導(dǎo)survivin ASODN轉(zhuǎn)染人膽囊癌細(xì)胞株GBC-SD,采用流式細(xì)胞儀技術(shù)檢查細(xì)胞凋亡變化,采用電鏡技術(shù)觀察細(xì)胞的形態(tài)變化,采用逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)檢測survivin基因表達(dá)的變化。
結(jié)果脂質(zhì)體介導(dǎo)轉(zhuǎn)染survivin ASODN后,膽囊癌細(xì)胞內(nèi)survivin mRNA表達(dá)相對系數(shù)為0.512±0.011,明顯低于未轉(zhuǎn)染者的0.980±0.012 (P<0.01),平均下調(diào)了47.83%(P<0.01),凋亡細(xì)胞比率為(26.28±3.91)%,明顯高于未轉(zhuǎn)染者的(0.50±0.23)%,P<0.01,電鏡下可見膽囊癌細(xì)胞呈典型凋亡樣改變。
結(jié)論survivin ASODN轉(zhuǎn)染后能下調(diào)survivin的表達(dá),可有效誘導(dǎo)GBC-SD細(xì)胞凋亡。
