【摘要】目的探討肝癌、癌旁及正常肝組織中Fas和FasL mRNA表達(dá)水平的變化與肝癌發(fā)生、發(fā)展的關(guān)系及其意義。
方法應(yīng)用半定量RT-PCR的方法檢測(cè)肝癌(40例)、癌旁(40例)及正常肝(25例)組織中Fas及FasL mRNA表達(dá)的相對(duì)含量。
結(jié)果正常肝組織、癌旁及肝癌組織中Fas和FasL mRNA表達(dá)的相對(duì)含量分別為0.792±0.039和0.245±0.043、0.857±0.031和0.429±0.035以及0.473±0.047和0.185±0.041。肝癌組織中Fas mRNA表達(dá)的相對(duì)含量明顯低于正常肝組織和癌旁組織(P<0.05); 肝癌組織中FasL mRNA表達(dá)的相對(duì)含量亦明顯低于正常肝組織(P<0.05)和癌旁組織(P<0.01),但癌旁組織中FasL mRNA表達(dá)的相對(duì)含量高于正常肝組織中的表達(dá)(P<0.05)。
結(jié)論肝癌組織不僅通過(guò)低表達(dá)Fas來(lái)逃避機(jī)體的免疫監(jiān)視作用,而且還通過(guò)癌旁組織FasL的高表達(dá),使與之接觸的淋巴細(xì)胞(表達(dá)Fas)凋亡,最終使肝癌細(xì)胞逃脫機(jī)體的免疫殺傷作用,得以增殖和轉(zhuǎn)移。
