【摘要】目的 探討保存溫度對(duì)移植胰腺功能的影響,確定大鼠胰腺的最適保存溫度。
方法 采用原位灌注、單純低溫保存及建立同系大鼠異位全胰十二指腸移植模型,按不同保存溫度(0 ℃、4 ℃、8 ℃、12 ℃)進(jìn)行分組,保存6 h后進(jìn)行移植,通過光鏡觀察胰腺組織結(jié)構(gòu)變化,高壓液相色譜法檢測(cè)保存6 h后移植物ATP和總腺苷核苷酸(TAN)含量。移植24 h后,檢測(cè)血糖、血清中脂肪酶及淀粉酶含量,測(cè)定移植胰腺組織中髓過氧化酶(MPO)活性,并進(jìn)行組織學(xué)觀察。
結(jié)果 4 ℃組保存6 h后和移植24 h后的胰腺組織結(jié)構(gòu)損傷明顯輕于其它組。保存6 h后胰腺組織中ATP和TAN水平在4 ℃組、0 ℃組、8 ℃組和12 ℃組依次降低,差異有顯著性意義(P<0.05)。移植24 h后血糖、血清脂肪酶及胰腺組織中MPO水平在4 ℃組、0 ℃組、8 ℃組和12 ℃組依次增高,差異均有顯著性意義(P<0.05)。結(jié)論4 ℃對(duì)大鼠胰腺是最適保存溫度,它能夠明顯減輕大鼠胰腺的冷損傷程度。
