摘要】目的 研究人白介素-13(hIL-13)對(duì)牛主動(dòng)脈內(nèi)皮細(xì)胞(BAECs)表達(dá)細(xì)胞間黏附分子-1(ICAM-1)和E-選擇素的影響,為hIL-13應(yīng)用于誘導(dǎo)異種移植免疫耐受、減輕異種移植排斥反應(yīng)提供實(shí)驗(yàn)依據(jù)。
方法 用不同濃度的hIL-13(2、5、10、20及40 ng/ml)孵育 BAECs 2 h后,再與腫瘤壞死因子-α(TNF-α, 4 ng/ml)共孵育6 h或18 h; 應(yīng)用細(xì)胞酶聯(lián)免疫吸附分析方法(Cell-ELISA)檢測BAECs表面的E-選擇素和ICAM-1的表達(dá); 用MTT方法測定hIL-13對(duì)BAECs活性的影響。
結(jié)果 用hIL-13在5~20 ng/ml濃度內(nèi)預(yù)處理BAECs后,能明顯抑制TNF-α誘導(dǎo)的E-選擇素與ICAM-1的表達(dá),并呈現(xiàn)一定的劑量依賴性(P<0.01)。經(jīng)不同濃度hIL-13處理的BAECs活性與未經(jīng)hIL-13處理者相比差異無統(tǒng)計(jì)學(xué)意義。
結(jié)論 hIL-13能明顯抑制BAECs表達(dá)E-選擇素與ICAM-1,誘導(dǎo)異種移植的免疫耐受,且hIL-13對(duì)BAECs無損傷作用。
